Connecting the PS555DR to the N272 streamer/preamp with a Burndy cable: is the cable directional? If so how is it determined?

Yes, you connect via an XPS or S-XPS Burndy and the band is what determines direction, with it going closest to the NAC-N272.
In the above image taken from the FAQ post about Burndy cables you can see the band Iām referring to on the XPS and the S-XPS Burndy cables.
